Effects of Corn-Derived Arabinoxylan-Containing Polysaccharides on Innate Immune Activity

Abstract

|
A water-soluble polysaccharide fraction derived from corn steep liquor, called corn byproduct polysaccharide-1-supernatant (CBP-1S), was analyzed for its immune activity in innate immune cells. Murine peritoneal macrophages, Raw264.7 cells, were treated with CBP-1S, which induced nitric oxide production in a dose dependent manner. The CBP-1S treatment also increased the inducible isoform of nitric oxide synthase and cyclooxygenase-2 as well as cytokine gene expression involved in the innate immune response. In addition, the proportion of dendritic cells expressing interleukin-12 was higher in the splenocytes of mice administered CBP-1S orally, which is expected to play an important role in the natural killer and T cell activity that inhibits tumor proliferation and metastasis. Arabinoxylan, a major component of CBP-1S, induced the phosphorylation of mitogen-activated protein kinase and promoted transcriptional activity through nuclear factor-kB. Therefore, this compound is believed to mediate signal transduction through the toll-like receptor 4 receptor. These results suggest the possibility of using corn-derived polysaccharides containing arabinoxylan as a functional food ingredient for improving the immune function.
